Thermostable DNA-ligase from Thermophilus aquaticus

Taq DNA Ligase is athermostable NAD+ dependent DNA ligase obtained from E. Coli cells, carrying plasmid with the corresponding gene Thermus aquaticus HB8.

The ferment catalyzes the formation of phosphodiester bond in duplex DNA between 5`- phosphate and 3`- hydroxyl groups, using NAD+ as a co-factor. For the process of DNA chain ligation, oligonucleotides have to be well coupled with the complementary DNA chain and have no gaps.

Taq DNA Ligase works at the temperature 45 - 70 °C.

Application:

  • Аllele-specific gene typing through Ligase Detection Reaction (LDR) and Ligase Chain Reaction (LCR)
  • Mutagenesis with the help of including phosphorylated oligonucleotide during the primer extension amplification.
  • Gibson assembly.
  • High-fidelity gene synthesis.
