TEV-protease

TEV protease is a highly specific cysteine protease of the Tobacco Etch Virus. The ferment is active in the wide рН range (from 4 to 8,5) and temperature range (from 4 ˚С to 34˚С). TEV protease recognizes and deconjugates the linear epitope, consisting of 7 amino acids. The preferential sequence for proteolysis ENLYFQ↓S(G), where ↓ means the broken down peptide bond.

This product is a genetically modified recombinant TEV protease, with molecular weight 28 kDa, containing at the N-end the sequence made up of 6 histidines, which allows to remove the ferment from the reaction mixture with the help of nickel chelate chromatography. 3 mutations were introduced into the coding sequence: S219V for the increase of the protein stability, and two (L56V and S135G) for increasing solubility.

The ferment was obtained from E. Сoli producer strain, affinity purified (the purity not less than 95 %) and transferred into the buffer for storage: 100 мМ Tris pH 8,0; 100 mM NaCl; 0,5 mM EDTA, 5 mM DTT, 50 % glycerol.
